Why does my upper back throttle shoulder hurt when I'm riding a sportbike?
Is it my positioning or because of the weight being supported by my arms?
I can't ride for more than one hour and then the pain becomes unbearable.

You shouldnt' be supporting your weight by your arms.

If your shoulder or wrists are hurting, you're gripping the bars too hard. Hold on with your body. Your hands should only be lightly on the bars (except for wheelies, stoppies, etc).

Toes on the pegs, and almost no pressure on the grips, use your knees and your perched position on the pegs to hold you on.

When I first got into sportbikes, my wrists were always killing me. Learned that at the track and haven't had a problem since.

It probably depends on the person. I rode my TT600 for around 700 miles in a day without problems. 400 miles was pretty routine on that bike. The 675 is a bit more extreme, but I have put in well over 100 on it and felt fine. It also depends on the kind of riding you are doing. If you are just droning down the freeway, then a sportbike is not ideal.

Yeah, you can ride all day like that, you find a comfortable position in the middle, and still keep the weight off your wrists and more importantly the pointy parts of your butt and tailbone.

It takes some doing to break the old habits, but once you learn the new ones, it's actually difficult to go back. I can ride all day, freeway, backroads stop and go, whatever... whereas it used to ache after 20-30 minutes.

Now, if you've recently dislocated your shoulder, or otherwise have some nagging problem inside of your body, this may not be as useful suggestions as they were for me.

What's a mean streak anyway?

My knees have a hard time with any distance. But by periodically flexing the legs I do fine. My ride yesterday was the first 100 miles + since surgery in Feb and it was painful after about 15-20 min. Maybe lower pegs would help, who knows.

Yeah, just try and use your torso & legs more to support your weight & it'll go away. I personally revisited this topic myself with my RC51 which felt like a torture rack designed to cripple my wrists for the first few days with it's even lower clip ons, but now I got used to it once my back/torso became conditioned to it. And as a bonus the S4 seat/bar orientation almost feel like a crusier or dirt bike!

Yeah, that's the same way my Buell feels when I pull it out of the garage. More like a flat tracker or motard with the bars. I'm half tempted to lean back and throw my foot into the turn.
