What did I mess up?

I did something stupid and need some opinions on what I may have caused.

I left the petcock in the prime position for about 100 miles. When I went to ride a few days ago, it barely starts. It takes a full choke just to turn over and sounds like it's missing badly, possibly running on only two cylinders. If I give it the least bit of throttle, it dies.

Is it possible I jacked up the plugs? The carbs were rebuilt about three weeks ago and it's been running strong, like it always has. This change came about suddenly.

I was out there to take her apart just a few moments ago and when I started to take the tank off to check the coils, I noticed the petcock position.

Normally, leaving the petcock in prime position wouldn't have much of an affect unless you have leaking float needle valves and seats. Did you renew the needle valve seats when you were in the carbs? I think that you're on the right track, look at the plugs first and see what they tell you. Also, check your oil for gasoline. If the float valves leak, gasoline often finds its way to the sump. Your carbs are Mikunis, right?

Only a thorough cleaning on the carb rebuild. Yes on the Mikunis. I'll take her apart and have a look. I'll check the oil also.

It's a pretty good bet that your float valve seats have cracked, the age is about right.

and that would splain why it wouldn't crank cause it was flooded
