Only 3 things are needed to make an engine run - fuel, compression and ignition. But timing for each is pretty important. Make sure you're getting fuel, not just to your Mikunis, but into the cylinder. After 1000 kicks (!), your plugs should be pretty wet (you might make sure you haven't fouled them with fuel). Then, since you've probably just reassembled your engine, make sure that both your valve and ignition timing are right - a spark arriving when there's no compression will get you nowhere.
