The carbs do not have to come off the bike to re-jet, it's done through the float bowls. However, you may have to remove the carbs to get some of those crappy stock carb screws undone (I did....I needed vice-grips).

Before you re-jet, make sure you get the stainless steel carb screws from BC or NB. The stock screws will basically self destruct upon removal.

Let's put it this way. Your bike won't blow up if you ride it around the block to see how your pipes sound. But you should re-jet as soon as possible. Don't go and put 300 miles on the bike this weekend while waiting for the screws to come in. I doubt if it will do any long term damage, but why risk it?

As far as the mixture screw, no, it's on the bottom of the carbs. The 865cc engines have a "D" shaped screw that does not need to be drilled out. You can either buy a d-tool from NB, or make your own tool from a .22 shell. I used plastic wire crimp connectors remove the screws, then sawed a slot into them so I could use a flathead screwdriver.

Check out these two guides:

ok , so i suceeded putting in the 4 jets , only mildly begining to strip 1 screw head. took her for a ride and was happy, until i hit 5500 rpm
and it hesitated and kinda burbled. it didnt do this before i did the jets.
i have preds and ai removal
did i neglect to do something in there or does it sound like the other high end jets might have been better, and if so ,which ones were the high end jets? thanks alot!

retro 9000, the main jets are the ones that you unscrewed out of the hex holder and the pilot jets were the ones that screwed out from the recessed hole. It sounds like you might be running a main jet that is to small. What size did you use? Did you do anything to the airbox like a snorkle removal? I don't know about the predetors, but I have the TOR's with the snorkle out. I'm using 130 mains and 48 pilot with the air screw out 1/2 turn Runs like a dream and really pulls off the line. If you don't know what size you have, the mains have it stamped on the side and the pilots are stamped on the rim.
